5-hydroxy-1,5-dihydro-2H-pyrrol-2-ones (5HP2Os) are next-generation thiol-reactive conjugating groups superior in reactivity and stability to conventional maleimides. Maleimides are prone to hydrolysis, retro-Michael reactions and/or thiol exchange reactions, resulting in antibody and protein conjugates with poor stability. 5HP2O linkers are highly selective for cysteines, yielding conjugates with superior stability.
