Sec allows the site-specific covalent protein modification, even in the presence of native cysteine, as the reactivity of a selenol group exceeds that of a thiol group. This photocaged selenocysteine (PSc) is incorporated as an unnatural amino acid in response to an amber stop codon using a mutant pyrrolysyl-tRNA/tRNA synthetase pair. Decaging can be achieved by irradiation with UV light (365 nm).
